ProATM ProATM is a new protein A chromatography resin that is generally used for purification of the various antibody proteins including monoclonal antibodies, polyclonal antibodies and Fc fusion proteins. Protein A ligand of ProATM is the native protein A originated from Staphylococcus aureus. Generally protein A is a surface protein found in the cell walls of the bacterium S. aureus. Since it is able to bind specifically to an antibody protein from many species of mammal, it has been exploited as an affinity ligand for the purification of recombinant proteins with antibody proteins. Protein A consists of five antibody-binding domain (E, D, A, B, C), and has a molecular weight of approximately 34.4 kDa. It is produced as recombinant proteins in E. coli through the high yield purification process. Protein A is coupled to the epoxy activated 4% highly cross linked agarose bead. The epoxy group has a feature of attaching to hydroxyl group (-OH), amino group (-NH2), or sulfhydryl group (-SH) under different pH condition. Especially, the binding capacity of protein A for antibody proteins is improved by coupling to agarose beads in the same direction.
